Should I build a small enclosure for 2 (one left and right) speakers in the back of my truck. Or is mounting them in a pannel (infinite baffle) good?

No they will be a 2 or 3 way component speaker. The signal to them is cut below 180hz.
I was kind of thinking of an enclosure. But I saw no recommendations by the manufacturer on making an enclosure. But I supose if an enclosure can enhance a sub it will do the same for mid and high range.1993 EXT. CAB, 3.4L V6 TBI, 5spd manual. Sonoma

Personally, I'd do infinite baffle for 6x9s, especially if you have a seperate sub. I actually would never run 6x9s, but that is just me. This of course coming from a guy who doesn't believe in rear fill and has over $1200 invested in front speakers alone.-Brad-

They will be 6.5 or 5.5 round speakers. Not 6x9's. But I supose in either case the same rule will apply.
The cut off it actually 120Hz I was wrong before.

Ok... So let's go over this. You will be using 6.5" or 5.5" speakers for midbass/midrange. Are they seperates or 2/3-ways? What are the other speakers in the system? It really depends on the rest of the system and what they will be used for when it comes to deciding how to enclose them. For instance, my front stage consists of a Dynaudio 7" midbass, a 4" Focal midrange and a 1" Dynaudio high. I am running the 7" in basically a freeair setup and they produce very clean, crisp bass all the way down to their cutoff at 60hz. The 4" take over at around 500hz and the 1" carry from ~2.5k up. I can run without subs (current config due to amp issues) and it covers the full audio spectrum pretty well, except for the vibrations from the lower bass.
Ok, so that doesn't really help you out any. But what I'm saying is, tell me what else you are running and where. If the speakers are going to be behind the seat and they are only for fill, then it will require a different setup than if they are to act as a main speaker.-Brad-

The will be in the back. They will be 2 way speakers. They will be cut off below 120hz and be expected to cover everything above. 2 12's handle the bass 120hz and below. 2 4x6 plate speakers do the work up front.

I would go with 6.5's and cut off at 100Hz, from the sound of it you can build a box for them if you like it may give you a bit better sound. But if you are not using components, don't go too crazy. Especialy if you are running 4x6's for your front stage. Plus, "infinate baffle" is something that more applies to subwoofers. You could try some of those prefab foam baffles too, but they are mostly a weather-proofing thing.
